  • Any proofreading polymerase amplified DNA (directly) or even Taq amplified DNA (with an additional blunting step),

  • With any top quality T4 DNA ligase,
  • Using a flexible ligation time from 5 minutes to overnight adjustable to your cloning needs a DNA that is an ORF or behaves as a bacterial promoter or produces a toxic gene can be cloned into pSpark®.

Introduction of pSpark® into a DNA cloning lab is seamless integrated and from the first time researchers find comfortable with kits because they are extremely user-friendly. After few uses any researcher becomes an advanced user of pSpark® DNA cloning systems.

All you need to begin to use pSpark® is to pipet specified amount of supplied reagents plus 5ng per kb of your insert and to incubate ligation at RT for 5 minutes to overnight (optimal ligation time of 60 minutes) and to transform all the resulting ligation mix into any competent E. coli.
